Cell Adaption Recommendations

New Life Biologicals recommends this approach for cell adaption.

Rapid Adaptation

  1. Grow cells up to a good confluence at around 1-2×106 in the typically used media with FBS at normal concentration. 
  2. Pass cells (reseed) into media containing the same percentage of CGP as the FBS inclusion level.

Extended Adaption

  1. Grow cells up to a good confluence at around 1-2 x106 in the typically used media with FBS at normal concentration.
  2. Pass cells (reseed) into media containing between 50-60% FBS and 40-50% CGP of the normal 100% inclusion level of FBS.
  3. Once cells have reached a good density, i.e. 1-2 x106, then pass into fresh media containing 25-30% FBS and 70-75% CGP of the normal 100% inclusion level of FBS.
  4. As the cells reach a good density, i.e. 1-2 x106, then pass cells (reseed) into media containing no FBS or if there is still growth lag to a lower FBS inclusion level.

Depending on the cell type and the media(s) being used, additional passages at the reduced FBS inclusion levels may be warranted during the process.

Also, depending upon cell type and media it has been demonstrated that additional supplementation in the media may be beneficial.

Notes:

  • Use of enzymes to release cells during adaptation is not recommended.
  • Better results have been achieved with mechanical release of cells from surfaces.
